本发明公开了一种用于辅助检测大豆百粒重的引物，包括如下一对引物：F：CGCCTTACTCTGTATCTTCT；R：CACCTCTTCCTCACTCCT。本发明还提供了一种检测大豆百粒重的方法，包括以下步骤：以待测大豆基因组DNA 为模板，将上述引物对上述DNA模板进行PCR 扩增；PCR扩增产物进行酶切，检测酶切产物中是否有238bp和181bp两条带，若酶切产物中有238bp和181bp两条带，上述待测大豆为大籽粒，若扩增产物中无238bp和181bp两条带，上述待测大豆为小籽粒。本发明的优点在于，可以通过对育种材料进行筛选，淘汰无238bp和181bp目标条带的亲本或育种中间材料，极大的提高了育种过程中选择高百粒重大豆品种的效率，对于高产大豆品种的培育效果显著。
